Hernan Organvidez, chief of the Doral police department, said police received a call at 1:30 am (0530 GMT) of "an active shooter and shots fired" at the resort.
Police officers from both the Doral and Miami-Dade departments responded and "confronted the subject immediately upon arrival and exchanged fire with him," he said. The shooter was "neutralized and taken into custody without further incident," he added.
Miami-Dade police said there were no shooting victims after the man opened fire in the resort lobby. One officer suffered a broken wrist.
The gunman was taken to a local hospital and his condition is unknown, police said.
"The incident took less than a couple of minutes," Organvidez said.
Eric Trump, a son of US President Donald Trump, thanked both police departments on Twitter, saying: "Every day they keep our community safe. We are very grateful to you!"
The 90-hole luxury resort in south Florida was purchased by Donald Trump in 2012 for 150 million dollars. Its Blue Monster Course is the site of PGA tour events.
