"The performance of 'Rusalka' was stopped just before the end of Act One following an accident on the stage involving Ana Maria Martinez," a Glyndebourne spokeswoman said.
"Ms Martinez fell into the orchestra pit. Despite Ana Maria wanting to continue with the performance, she was immediately taken to hospital as a precautionary measure.
"Her understudy, Natasha Jouhl, who plays the role of the First Nymph in the opera, continued in the principal role, while her understudy took the role of the First Nymph.
"Fortunately, no one else was hurt in the incident."
Martinez was "a little bruised" but "fine" and was sent home to recover, the spokeswoman added.
Among her string of accolades, Martinez sang alongside Placido Domingo on a recording of Isaac Albeniz's "Merlin", which won a Latin Grammy in 2001.
She was due to appear again on the Glyndebourne stage on Monday and on August 28.
In her next scheduled appearance following Glyndebourne, Martinez is due to perform in concert with Domingo in Caracas on September 2, followed by dates in Colombia and Peru.
